Madden was the first to strike, chipping home in the 34th minute after capitalising on some poor Brentford defending.
Upson then marked his 100th Yeovil appearance with a wonder strike in the 41st minute, letting rip from 30 yards and giving Bees goalkeeper Simon Moore no chance.
Prior to those two goals, it had been the visitors who looked far more dangerous.
Brentford could, and probably should, have been in front, only to be denied by the excellent Marek Stech.
The Yeovil keeper tipped Clayton Donaldson's speculative effort over the bar, and also made a crucial block with his knees to deny Adeyemi in the spell before Madden's opener.
Sponsors' man of the match Dan Burn made it 3-0 in the 84th minute with a cracking finish, hitting Jamie McAllister's free-kick first time.
And a few minutes later Brentford were lucky they didn't go four down when Kevin Dawson's effort was cleared off the line and Sam Foley fired just wide.
